چکیده

1. Plasma and buffy-coat vitamin C, urinary proline, hydroxyproline, creatinine and total amino acid concentrations were meausred in 23 healthy elderly subjects at intervals of 3 months. 2. There was a strong positive correlation between plasma vitamin C and buffy-coat vititamin C. 3. There were not significant correlations between plasma or buffy-coat vitamin CPAMIN C. 3. There were not significant correlations between plasma or buffy-coat vitamin C and total urinary hydroxyproline, whether expressed on a creatinine basis or on a total amino acid basis. Similarly, no significant correlations could be detected involving the proline/hydroxyproline ratio in urine hydrolysates. 4. There was a significant negative correlation between plasma or buffy-coat vitamin C and total urinary proline, when expressed per unit of total urinary proline, when expressed per unit of total urinary proline, when expressed per unit of total amino acids in the hydrolysates. This correlation was not observed with unhydrolysed urine, and it appeared to reside in the diffusible fraction, part of whose proline could be liberated by prolidase digestion. In addition, in the man, there was some evidence for a positive correlation between plasma or buffy-coat vitamin C and the ratio of total urinary amino acids to creatinine. 5. These results support the view that poor vitamin C status in elderly humans may be associated with a defect in collagen proline hydroxylation, reflected by increased excretion of proline-rich, collagen-derived peptides. If this interpretation is correct, it indicates a potential defect in connective tissue repair, and could form the basis of a functional test for subclinical vitamin C deficiency.

Prevalence of proline racemase/ hydroxyproline epimerase gene in human brucella isolates in Iran

Background: Human brucellosis is a zoonotic disease caused by Brucella melitensis, Brucella abortus, and Brucella suis. Brucella causes a chronic disease, which subverts the immune defense system of their hosts. In this study, the prevalence of an important Brucella virulence determinant, PrpA, which can modulate immune response, was determined in human isolates. Methods: Polymerase chain reac...

Induced connective tissue metabolism in vivo: reutilization of pre-existing collagen.

Light and electron microscopy of resorbing connective tissues have shown that the collagen fibers and fibrils disappear. It is usually presumed that resorbed collagen is degraded proteolytically to hydroxyproline-containing peptides and to free amino acids, including hydroxyproline. We wish to present evidence that collagen can be resorbed and then reutilized without such breakdown. Urinary exc...

Effect of acute aerobic activity after a period of taking vitamin C supplement on Iron status and hematological indices of professional football players

Vitamin C consumption is important in Iron absorption. Therefore, the purpose of this study was to investigate the effect of acute aerobic activity after two weeks vitamin C supplementation on Iron status and hematological indices in club football players. In this quasi-experimental study, 40 club football players from Khuzestan province were randomly divided into two groups of vitamin C and pl...

Urinary Hydroxyproline Excretion in Orthopedic Disease. Preliminary Report.

The significance of urinary hydroxyproline has not been completely clarified. It appears, however, that this amino acid excretion is correlated with either synthesis or destruction of collagen. Collagen hydroxyproline is reported to be formed by the irreversible hydroxylation of peptid-linked proline and no free hydroxyproline is incorporated directly into this protein. Therefore, the changes o...

The effect of tryptophan load and vitamin B 6 supplementation on urinary excretion of tryptophan metabolites in human male subjects.

I. Urinary excretion of tryptophan metabolites of the kynurenine pathway before and after a loading dose of 2 g of L-tryptophan (both before and during supplementation with vitamin B6) was studied in normal young male subjects and in elderly male subjects with prostatic diseases. 2. The increase in urinary excretion of tryptophan metabolites in the normal male subjects after a loading dose of 2...
